Editing with pspad (V4.5.6, on Windows 7) a 2.2 MB text file in my dropbox directory leads to an error when trying to save the file to the same location (pspad reports an error when saving the file).
Editing with pspad the _same_ text file in a directory outside dropbox causes no problem when saving it.

I don't know dropbox from the inside to answer you. I don't know, how DropBox is programmed, how he detects changes e.t.c.
Can you send me the error what you got or make a screenshot?

I am using PSPad 4.5.8 (developer build)
I installed dropbox, put 4MB file (english PSPad spell check dictionary) into dropbox folder. Open it in PSPad, modified and saved. I didn't meet any problem.
Can you download latest developer build, backup your current PSPad.exe and replace it with PSPad.exe from ZIP archive (developer build).
YOu can easy switch it back to your current version.

I tested with pspad V4.5.8 (build 2462).
I successfully was able to save my file (to my local dropbox folder) 2 or 3 times...only.
Next attempts to save have lead to the same error while saving with V4.5.6).
Note that with the V4.5.6 version I was able to "only" save my file once (the 1st time).
Can it be related to the sync process of dropbox operating on the file at that time?

Yes, it's possible it can depend of synchronization process. I guess if synchronization is running, the file is locked.
When you wait a while, is possible to save your file?

... makes sense.
But, even once the file is synced to "the cloud", I may not be able to save it again!
I filed a bug report at dropbox...
