PSPad uses Windows Scripting Host. You can install any language suitable for WSH.
PSPad will create folder for all languages supported by WSH.
In case PSPad is installed in program files, you need to start PSPad as admin to give it rights to write there.

I mean that create folder PSPad\Script\AutoIt.
Folder contains files "au3".

In each of these files have a line

Sub Init
addMenuItem "AutoIt.1","AutoIt", "AutoIt1", ""
End Sub

In these files, there is a function
Func AutoIt1()
Statements
EndFunc

No.
WSH is Windows scripting Host engine. It serves as host for another languages. Part of Windows is JScript and VBScript. You can install another languages like PHP, Perl, ...
I don't know if AutoIT suports to be hosted by WSH.
To create folders manually won't help you. PSPad gets list of languages from system and creates folders for languages which are installed when you start PSPad and when PSPad has rights to do it.
